DNMT1 or DNMT3a knockdown at late stage of the differentiation promotes lipogenesis in 3T3-L1 cells. DNMT1 or DNMT3a knockdown at the late stage of the differentiation promoted the expression of lipogenic markers such as Srebp1c (A) and its downstream target Fas (B), Acc (C) and Scd1 (D), and enhanced GPDG activity (E). 3T3-L1 cells at day 5 of the differentiation were electroporated with Dnmt1 or Dnmt3a siRNA and differentiated as described in Materials and Methods. Cells were harvested at day 8 of the differentiation for the measurement of gene expression. Gene expression was measured by real-time RT-PCR and normalized to cyclophilin.
