Regulation of Wnt target gene expression by USP6. (A) USP6 activates Wnt signaling in HeLa cells: HeLa cells were transfected with plasmids encoding USP6 and a Wnt/β-catenin reporter. The cells were incubated with indicated dilutions of WNT3A-conditioned media, and luciferase activity was measured after 24 h. (B and C) Activation of AXIN2 and NKD1 expression in HEK293 cells transfected with USP6: HEK293 cells were transfected with plasmids encoding full-length USP6 or its variants and incubated with WNT3A CM or control media for 24 h. AXIN2 and NKD1 expression was measured by quantitative RT-PCR (qRT-PCR). Data are presented as mean ± SD (n = 3).
