Screening of DG lipase activity. (A) DG lipase activity in the soluble fraction of CHO cells expressing the candidate proteins (100 µg) was measured using 40 µM cold SAG as a substrate. The assay mixture was incubated at 37°C for 30 min. Lipids were separated using TLC and visualized using iodine vapour. Lane 1: acyl-peptide hydrolase, lane 2: anti-depressant-related protein ARG123, lane 3: dihydropyrimidinase-like 2, lane 4: TUC-4b, lane 5: human DDHD2, lane 6: vector control, lane 7: 5 nmol 2-AG.
