Naïve CD4 T cells were sorted and cultured with WT antigen presenting cells, in the presence of anti-CD3 and anti-CD28. (A) Cell proliferation was measured by CellTrace Violet after 3 days of co-culture with WT APCs, in the presence of 0.1 ug/ml anti-CD3 and 3 ug/ml anti-CD28. The grey filled histogram represents control cells cultured with APCs but without anti-CD3 and anti-CD28, for each genotype. (B) IL-4 production was measured by restimulating and intracellular staining of cells after 3 days of co-culture with WT APCs, in the presence of 3 ug/ml anti-CD28 and varying concentrations of anti-CD3. Representative data from 1 of 2 independent experiments is shown.
