Design and cloning of SDF1-ELP. (A) Cloning of SDF-ELP was done using a peT25B+ expression vector. SDF1 was fused to ELP using the XbaI and NdeI restriction sites. The plasmid with SDF1-ELP was mutated to put a 6X Histidine tag in frame with the protein sequence. The plasmid diagram was obtained using SnapGene® software (from GSL Biotech; available at snapgene.com). (B) Pymol rendition of SDF1-ELP. SDF1 (in yellow) is separated from ELP (red) by a linker (in blue) comprising 3 repeats of 4 glycines and 1 serine. SDF1 monomer sequence was extracted from Ryu et al.[30] (RSCB Protein Data Bank ID: 2J7Z). The ELP portion was modeled using I-TASSER software [31],[32],[33].
