SphK activity is increased in response to various stimuli such as growth factors, which results in intracellular generation of S1P.

Notes: After its release in the extracellular space, S1P can activate its receptors and promotes tumor progression, angiogenesis, invasion, and metastasis of HNSCC. Various components of S1P signaling pathway, such as S1P, SphK, and S1PRs, are potential drug targets for cancer.

Abbreviations: SphK, sphingosine kinase; S1P, sphingosine-1-phosphate; HNSCC, head and neck squamous cell carcinoma; S1PRs, S1P receptors.
