Hypothermic machine perfusion (HMP) is associated with less kidney inflammation than cold storage (CS). (A) Nuclear factor-κB (NF-κB) expression was evaluated by western blot analysis. The HMP group showed significantly decreased NF-κB expression compared with that in the CS group (**P<0.01). IκBα expression was evaluated by (B) western blot analysis and (E) RT-qPCR. The HMP groups showed significantly increased IκBα expression compared with that in the CS group (**P<0.01, *P<0.05). Tumor necrosis factor-α (TNF-α) expression was detected by (C) RT-qPCR and (D) ELISA. The HMP group showed significantly decreased TNF-α expression at both the protein and mRNA levels compared with that in the CS group (**P<0.01) whereas there was no significant difference in the expression of TNF-α between the HMP and sham groups (P>0.05). Results represent the means ± SD of three experiments.
