Role of caspase-3 in PES-induced apoptosis in U937 human leukemic cells. U937 cells were incubated at the indicated concentrations of PES with or without caspase-3 inhibitor z-DEVD-fmk (50 µM). (A) The growth inhibition of PES was suppressed by z-DEVD-fmk. (B) Cell cycle analysis. The accumulation of sub-G1 DNA content in cells was decreased by z-DEVD-fmk. (C) 4′,6-Diamidino-2-phenylindole (DAPI) staining. The number of apoptotic bodies was decreased by z-DEVD-fmk. Stained nuclei were observed under a fluorescence microscope using a blue filter (magnification, ×400). (D) DNA fragmentation test.
