The role of MAPKs in the PES-induced apoptosis of U937 cells. U937 cells were incubated at indicated concentrations of PES for 24 h. (A and B) Western blot analysis revealed that PES increased p-ERK and p-p38 MAPK in a dose- and time-dependent manner in U937 cells. (C–E) Cell cycle analysis shows the accumulation of subG1 DNA content. ERK inhibitor (PD98059) (PD) suppressed the PES-induced apoptosis, while JNK inhibitor (SP600125) (SP), and p38 inhibitor (SB203580) (SB) did not. The results are from one representative of three independent experiments that showed similar patterns. PES, polyphenols from Korean E. supina; MAPKs, mitogen-activated protein kinases.
