Effect of BEZ235 on HCT15 colorectal cancercells. (A) Cytotoxic effect of BEZ235 on HCT15 cells. Tumor cells were seeded at 5 ×10⁴ cells per well (0.5 mL) in 24-well culture plates, incubated for 24 h at 37°C and then treated with increasing doses of BEZ235 for 1d, 2d, and 3d. Cell viability was measured by MTT assay. The viability of the untreated cells was regarded as 100%. Experiments replicated three times. (B) Effect of BEZ235 on Akt and ERK activity in HCT15 cells. Cells were treated with BEZ235 at a concentratons ranging from 20 to 1,000 nM/L for 24 h. Cell were lysed and subjected to Western blot analysis using antibodies for p-ERK^Tyr204, ERK, pAkt^Ser473, Akt and β- actin. β -Actin served as loading control.
