The amount of hydroxyapatite was determined using an OsteoImage assay (Lonza, Walkersville, MD). The fluorescent staining reagent (green) binds to the hydroxyapatite portion of the mineralized matrix, and the fluorescence is measured at 495/519 nm (Ex/Em). The initial cell density was 1.6 × 106 cells/sample. (a) The images exhibited green fluorescence proportional to the amount of hydroxyapatite in the samples. (b) A progressive increase in the deposition of hydroxyapatite occurred in the paper scaffolds over time (n = 30). The scale bar represents 30 μm.
