The ΔrbdA and ΔdscE mutant strains are rescued for hypoxic growth by expression of cleaved N-SrbA but not full-length SrbA. AkuBKU80, ΔrbdA, and ΔdscE strains were transformed with plasmids expressing N-SrbA-myc (cleaved activated form) or full-length SrbA-myc under the endogenous srbA promoter. ΔrbdA-NsrbA-myc and ΔdscE-NsrbA-myc strains show partially restored growth under hypoxia, whereas ΔrbdA-srbA-myc and ΔdscE-srbA-myc strains do not, suggesting that RbdA and DscE are involved in srbAp activation by cleavage of its C-terminal domain.
