(b) pHA-PPARγ AF1 (3 μg), pHA-PPARγ DBD (4 μg) or pHA-PPARγ LBD (3 μg) was overexpressed with pFLAG-FBXO9 (1 μg) for 24 h and treated with MG132 (10 μM) for 5 h before harvesting. The deletion mutants of PPARγ were immunoprecipitated using anti-HA antibody-conjugated beads and immunoblotted using anti-FLAG and anti-HA antibodies. (c) GST-FBXO9 (1 μg) and PPARγ LBD (2 μg) were mixed and incubated. Co-precipitates were immunoblotted using anti-PPARγ or anti-FBXO9 antibodies.
