(A,B) Flounders were infected with megalocytivirus RBIV-C1 in the presence or absence (control) of pol-miR-731 agomir, pol-miR-731 antagomir, agomir negative control, or antagomir negative control, and the expression level of IFN in spleen was determined at mRNA and protein levels at 4 days post-infection (dpi) by qRT-PCR (A) and western blot (B), respectively. (C) mRNA levels of IFN-stimulated genes in the above samples were determined by qRT-PCR. (D) FG-9307 cells were transfected with or without (control) pGL3-IFNp plus pol-miR-731 mimic, mimic control, pol-miR-731 mimic + pCNIRF7, or pol-miR-731 + pCN3; the cells were treated with poly I:C, and luciferase activity was measured at 24 h post treatment. All experiments were perforemed three times, and values are shown as means ± SEM. *P < 0.05, **P < 0.01.
