D5D-KD promoted DGLA's anti cancer effect in colon cancer cells. (A) MTS assay of HCA-7 colony 29 and HT-29 cells treated by 1.0 µM 8-HOA; (B) Western blot and relative protein expression of D5D and COX-2 expression in HCA-7 colony 29 and HT-29 cell lines after D5D siRNA transfection (loading control: β-actin). The relative ratios of D5D or COX to β-actin for each cell line were normalized to 1 respectively; (C) MTS assay of HCA-7 colony 29 and HT-29 cell lines (control siRNA transfected vs. D5D-KD) after 48 h of DGLA or AA treatment (100 µM).
