Figure 1. Different modes of lipid accumulation.
Examples of CARS images of control cells (no treatment) (a), cells treated with Entinostat (b), cells treated with oleic acid overload (c), cells treated with both oleic acid overload and Entinostat (d). Entire fields of view are shown on the left, and insets with cell outlines overlaid on the right. Entinostat induces a moderate degree of lipid droplet accumulation across almost all cells (b), whereas with oleate (free fatty acid), there is significant increase in lipid accumulation in some cells, observed as higher numbers of lipid droplets per cell as well as larger volumes of individual droplets; two examples of different lipid structure accumulation are shown in the expansion in (c). Entinostat treatment under conditions of excess metabolite availability results in a strong lipid loading in almost all cells (d). Images are single planes taken from z-stacks, displayed after noise removal using PureDenoise (see Materials and Methods). Scale bars for images are 30 μm and 10 μm for expansions.
