(A) Halo-TAX1BP1, mCherry-OPTN, and GFP-NDP52 colocalize on depolarized mitochondria 90 min post-CCCP in Parkin-expressing HeLa cells. (B and C) Recruitment of either NDP52 or TAX1BP1 to depolarized mitochondria is dependent on Parkin and blocked by expression of the Parkinson’s disease-associated Parkin-T240R mutant. HeLa cells transfected with Mito-DsRed2, mCherry-Parkin-T240R, and either Halo-NDP52 (B) or Halo-TAX1BP1 (C). At 60 min post-CCCP, Parkin-T240R is recruited to depolarized mitochondria, but NDP52 and TAX1BP1 remain largely cytosolic.
