Monosaccharide O-fucose and terminal dixylose of O-glucose-linked saccharides play redundant roles in relocating Notch from the apical plasma membrane to AJs. A–I‴, wing discs with somatic clones homozygous for O-fut1R245A (A–C‴) and shams (D–F‴) or double-homozygous for O-fut1R245A and shams (G–I‴) were stained with antibodies against NECD (A′, A″, D′, D″, G′, and G″); NICD (B′, B‴, C′, C‴, E′, E‴, F′, F‴, H′, H‴, I′, and I‴); DE-cad (B″, E″, and H″); and PDI (C″, C‴, F″, F‴, I″, and I‴).
