Analysis of simulated single channel currents for the adult human wild-type AChR activated by ACh. For each of the indicated ACh concentrations, a segment of simulated single channel activity is displayed at a bandwidth of 25 kHz (Materials and methods), and the corresponding closed and open time histograms are displayed with the global fit of Scheme 1 to the data overlaid. Rate constants for simulation are those from the global fit of Scheme 1 to the experimental data (Table 1), but with channel gating rate constants of β₂ = 125,000 s⁻¹ and α₂ = 2100 s⁻¹ determined by iterative simulations followed by kinetic fitting (see Table S4). Rate constants from fitting Scheme 1 to the simulated data are presented in Table 1.
