Effects of Mg^II concentrations on the TdT-catalyzed polymerization of (a) natural thymidine triphosphate (dTTP) and (b) artificial dHTP. Primer (5′-FAM-dT₂₀-3′): 5.0 µM, dTTP or dHTP: 100 µM (20 equivalents), TdT: 2 U/µL, MgCl₂: 0, 1.0, 2.0, 5.0, 10, 20, and 50 mM, 37 °C, 24 h. The products were analyzed by denaturing polyacrylamide gel electrophoresis (PAGE). The bands were detected by fluorescence of the fluorescein (FAM) moiety. “n” represents the number of the appended nucleotides. “nt” is short for “nucleotide”.
