V-ATPase is composed of a peripheral sector V₁ (green), which carries the ATPase activity, and the proton-conducting, membrane-integral V₀ sector (red and yellow). Only the V₁ and V₀ sector subunits used in this work are indicated. The black star in the V₀ a-subunit Vph1 represents the residue R735. Glucose withdrawal releases V₁ from V₀ and renders it soluble in the cytosol. Proton conductance by V₀ is blocked in this state.
