PBISe induces caspase dependent apoptosis of human HCC in a dose-dependent fashion. Four different cell lines were used to demonstrate the apoptotic process. (A) Skhep1; (B) HepG2; (C) C3A; (D) Huh-7. HCC cells were seeded onto 96-well plates (5 × 103/well) in MEM with 10% FBS and allowed to attach overnight. Cells were then treated with PBISe or DMSO at indicated concentrations the following day, as shown in the figure. After 48 h of treatment, cell apoptosis assays were performed using Apo-one Homogeneous Caspase-3/7 Assay (Promega) according to the manufacturer’s instructions. n = 3; error bar represent Mean ± S.E. The protein expression of cleaved PARP analysis was performed by Western blot analysis as shown in figure for all cell lines.
