HDAC8 inhibition does not alter regulation of cohesin-sensitive ER target genes. Estrogen-starved MCF7 cells were transfected with control non-targeting or RAD21 siRNA (A) or SMC3 siRNA (B), or were treated with 10 or 25 μm of PCI-34051 (C) for 48 h, then treated with 100 nm estradiol for 6 h. Transcript levels of the indicated genes were quantitated by qRT-PCR and are expressed relative to the mean of two reference genes, CYCLOPHILIN and GAPDH. Error bars represent the mean ± S.E. of three independent biological replicates for each treatment type.
