Partial rescue of FAO in mdh3Δ mutant by PXN. A, Oleate-inducible expression of PXN (pHHU274; 36.1 kD) in the mdh3Δ mutant background was confirmed by immunoblot using PXN-specific antibody. Total yeast membrane proteins isolated from the wild type and different yeast mutants were analyzed. B, Growth curves of wild-type and mutant strains on oleate (C18:1) medium as sole carbon source. The strains shown are: wild-type cells (▪), mdh3Δ cells transformed with empty vector (●), mdh3Δ cells expressing Mdh3p (▴ in gray), and mdh3Δ cells expressing PXN (♦). As negative control fox1Δ cells (○) were used. C, Wild-type and mdh3Δ cells transformed with the empty vector or expressing PXN or Mdh3p, grown on oleate medium, were incubated with [1-14C]-octanoate and β-oxidation activity was measured.
