Determination of putative glycosylated proteins by label-free proteomics and enrichment by ConA chromatography. A, Normalized abundances obtained by label-free analyses of trypsin digests of crude proteins and proteins eluted from ConA columns were used to classify proteins (with an ANOVA p value < 0.05) from the two data sets by the AutoClass Bayesian clustering system (webserver Autoclass@ijm). A higher abundance of the protein in the eluate from the ConA column than in the crude sample was considered to indicate that the protein was probably glycosylated. B, Putative glycosylated proteins from clusters defined in (A) with higher abundances in mca1Δ/Δ-F conditions.
