MCAS (A) and RMUG-S (B) cells (1 × 10⁴) expressing sh-luc, sh-CASZ1 (2), or sh-CASZ1 (3) were seeded in 12-well plates and incubated for 4 days. The number of cells was counted each day. Each data point represents the mean ± SEM of 3 wells. (C, D) The effect of CASZ1 knockdown on anchorage-independent growth in MCAS (C) and RMUG-S cells (D). Briefly, 2 × 10⁴ of the indicated cells were plated in soft agar, incubated for 4 weeks, and then stained with 0.01% crystal violet. The number of visible colonies was counted.
