S. aureus and SEA-induced IFN-γ responses are partly IL-12-dependent. PBMC were cultured in the presence of an IL-12 neutralizing or isotype antibody, stimulated as indicated, and analyzed by flow cytometry. The relative percentage of IFN-γ+ cells among the CD4+ (n = 7), CD8+ (n = 7), γδ TCR+ (n = 7), CD161+Vα7.2+ (n = 4), and CD3CD56+ (n = 7) lymphocyte populations is shown. The percentage of IFN-γ+ cells in isotype-treated PBMC stimulated with S. aureus or SEA is set to 1, and the percentage of IFN-γ+ cells in α-IL-12-treated cultures is relative to the isotype. Bars show medians with interquartile range.
