A, B. Cytotoxicity assay to assess the PBMC/NK92 cell-mediated killing of MDA-MB-231 cells. The EC50 of mAb04-MICA/mAb04 were fitted. C, D. MDA-MB-231 cells were used as target cells and PBMC/NK92 cells as effect cells for a LDH release assay. The increase of cell lysis was enhanced significantly upon treatment with mAb04-MICA compared to mAb04. Measurements were performed in triplicates (A-D). E. MDA-MB-231 cells were incubated in triplicate with mAb04-MICA/mAb04/AK404R and rabbit serum. Following washing, cells were stained with PI and analyzed by flow cytometry to assess viability. Data were expressed as the mean ± SD of PI-positive percentage.
