Effect of Listeriolysin O (LLO) on luciferase (Luc) activity. Luciferase activity (relative light unit (RLU) × 105 units per μg protein) expressed in Her18 versus MCF7 cells is plotted against dose (5 to 15 μl) of DNA complexes used for treatment. Cells were treated with DNA complexes containing trastuzumab (Tmab)/Luc and with or without LLO. Each treatment was set up in triplicate, and cells were harvested 24 h after treatment for analysis of luciferase activity. Bicinchoninic acid (BCA) protein determinations were done on the same cell extracts used for the luciferase assay. The data are shown as mean±s.e. (n=3).
