Insulin signaling blockade in transplanted endodermal committed blastomeres promotes Pdx1 expression. (A) Scheme of endoderm replacement transplantation experiments. sox32-injected blastomeres termed “Endoderm Committed Blastomeres” (ECBs) were isolated from donor Tg(sox17:GFP) blastulae with or without injected dnIRS2-GFP mRNA. ECBs were transplanted into endoderm deficient, sox32MO-injected host blastulae. (B) 24 hpf chimera shows extensive replacement of endogenous endoderm with transplanted Tg(sox17:GFP) donor cells.
