Objective-lens based TIRF microscopy to detect specific interactions between flurophore-free α-Syn monomers on the cover slip and fluorophore-labeled counterparts in solution. The experiments are carried out at acidic pH. A sudden increase and abrupt drop of fluorescence intensity are unambiguously ascribed to the association and dissociation of a dimer within the evanescent field of TIRF, respectively. The short dwell time between association and dissociation are interpreted as a lifetime of a dimer.
