LAMP-3 expression is controlled at the transcription and translation level by LPS and Salmonella in THP-1 and HeLa cells. THP-1 cells, at a density of 1.0 × 106 cells/well, were treated with 1 ug/ml of LPS (A, B) or infected with S. typhimurium (C, D), or HeLa cells were infected with S. typhimurium (E, F). (A, C, and E) At each time point, total cellular RNA was extracted and subjected to RT-PCR with gene specific primers. Tubulin gene was used as an internal control. (B, D, and F) Protein levels of LAMP-1, LAMP-2 and LAMP-3 were examined by Western blotting with specific antibodies. GAPDH was included as a loading control.
