Role of V-ATPase in the early response to ethanol-induced cell wall stress. The exponentially growing wild-type (BY4742) (■), Δvma2 (□), and Δvma3 (△) cells were incubated in YPD medium lacking ethanol or in YPD medium containing 5% or 10% ethanol at 30°C for 1 and 3 h. Cells were harvested and adjusted to an OD₆₀₀ of 0.5 in TE buffer containing 100 μg/ml (1 U) Zymolyase 20T. Susceptibility to Zymolyase was monitored by measuring the OD₆₀₀ at the indicated times and expressed as a percentage of OD₆₀₀ relative to that at 0 min. All experiments were carried out in triplicate. The error bars indicate the SD.
