Human DCs transfected with control siRNA or IFN-λR1 siRNA for 24 h were infected by mock or DENV for an additional 8 h. Cells were collected for measurement of NF-κB (A), AP-1 (B) or SP-1 (C) DNA-binding activity by EMSA or COX-2 expression by Western blotting (D). The NF-κB DNA-binding activity of DCs treated with or without IFN-λ1 was determined (E). The results pooled from at least three independent experiments using different donor cells are presented next to the representative figures.
