Wild-type and TLR7/9-/- BMDCs were cocultured with heat-killed or live Histoplasma capsulatum (HK-Hc or Hc, respectively) at an MOI of 2. One hour post infection spleen-purified native or Histoplasma-primed CD4+ T cells were added to the BMDCs. Supernatants were collected at 48 hpi and IFNγ was quantified by ELISA.
