Farnesyl diphosphate cyclization reaction catalyzed by pentalenene synthase from Streptomyces exfoliatus UC5319, as initially proposed by Cane and colleagues.18 This reaction is the first step in the biosynthesis of the antibiotic pentalenolactone. The co-product inorganic pyrophosphate (PPO, i.e., the PPi anion) is proposed to be the general base-general acid that mediates stereospecific proton transfers in catalysis.
