ROS production in SOD2 overexpressing human ASC after acute hypoxic challenge (1 mM H2O2 for 30 min) assessed using (A) the CellROX oxidative stress fluorogenic probe (original magnification 40×) and (B) fluorescence quantification with ImageJ software. Cell viability assay after 24 h of culture in cobalt chloride-induced hypoxic conditions (C). Treatment with the antioxidant scavenger N-acetylcysteine (NaC) (2 mM) was used as control. Results are reported as mean ± S.E.M. of three independent experiments. Significance was assessed by Mann–Whitney U test; * p < 0.05 versus ASC mock.
