Epicutaneous CpG ODN enhances Th1 responses. C57BL/6 mice adoptively transferred with CD4+ Thy1.1+ OT-II T cells were immunized with scOVA ± ecCpG or scCpG ODN. Skin draining lymph nodes and spleen were collected 3 days after and cells were restimulated with PMA and ionomycin ex vivo for 4 h. Cells producing IFN-γ were detected by intracellular IFN-γ staining and flow cytometric analysis. (A) Representative flow cytometric plots of CD4+ and Thy1.1+ cells, gating for CD4 IFN-γ double-positive cells is shown.
