PanP-DM1 retains the property of PanP. (A) ELISA evaluating the masked binding activity of PanP-DM1 to immobilized EGFR. EC50 for Pan, Pan-DM1, PanP and PanP-DM1 were 0.1265 µg/mL (95% confidence interval [CI], 0.1089–0.1469 µg/mL), 0.2034 µg/mL (95% CI, 0.1805–0.2293 µg/mL), 2.257 µg/mL (95% CI, 1.861–2.739 µg/mL) and 2.569 µg/mL (95% CI, 2.044–3.229 µg/mL), respectively. (B) FACS assay assessing the binding activity of activated PanP-DM1 (Ac PanP-DM1) to EGFR expressed on surface of A431 cells at 1 µg/ml or 10 µg/ml. (C) uPA-activated PanP-DM1 restored binding comparable to Pan-DM1, as tested by ELISA. EC50 for activated PanP-DM1, PanP-DM1 and Pan-DM1 were 0.3198 µg/mL (95% CI, 0.2804 - 0.3648 µg/mL), 2.414 µg/mL (95% CI, 1.950 – 2.987 µg/mL) and 0.2251 (95% CI, 0.1964 – 0.2579 µg/mL), respectively.
