T-jump (=7 °C) induced fluorescence (λ_ex = 295 nm and λ_em = 355 nm) time courses as a function of final temperature for the W87F form of ht-ADH. The plotted intensities were normalized to “probe-only” emission spectra obtained at −0.6 μs, and the experimental data were fit to a double exponential (Table 1). The final level of protein fluorescence is within 4% of the initial fluorescence in each case.
