Figure 4

Effects of sex steroids on major TACC2 isoform expression in breast carcinoma cells. (A, B) Effects of DHT on the TACC2 expression in MCF‐7 (A) and MDA‐MB‐453 (B) cells, and (C) Effects of estradiol on the TACC2 expression in MCF‐7 cells. These cells were treated with indicated concentration of dihydrotestosterone (DHT) or estradiol for 24 h. The mRNA level of TACC2 major isoform was examined by real‐time PCR analysis (upper panels). TACC2 mRNA level was evaluated as a ratio of RPL13A mRNA level, and subsequently relative TACC2 mRNA level was summarized as a ratio (%) compared with the basal level (nontreatment). Data are presented as the mean ±S.D. (n = 3). The statistical analyses were performed using one‐way ANOVA and Fisher's protected least significant difference. Lower panels summarized the TACC2 protein level by immunoblotting. 10 μg of protein was loaded in each lane, and β‐actin immunoreactivity was shown as the internal control.
