(a) CD4 and CD8 T cells were purified from NOD.ABD and control B10.H2g7 splenocytes as described in Materials and Methods, and 1×105 cells cultured with or without CD3 and CD28 antibodies at 37°C for 3 days. Concentrations of IFN-γ and TNF-α were determined by CBA (see Materials and Methods). Data represent the average values of triplicate samples ± SD and were consistent in two repetitions. (*, p<0.05, NOD.ABD vs. B10.H2g7 ). (b,c) Liver lymphoid cells were isolated and stained intracellularly as described in the Materials and Methods. The frequency of IFN-γ positive cells in CD4+TCRβ+ and CD8+TCRβ+ cell populations were analyzed by flow cytometry. One representative example of IFN-γ in CD4+ and CD8+ T cells is shown in (b).
