Female 6- to 8-week-old mice (n = 5 per group) were randomized to treatment with vehicle or compound by i.p. administration followed by inoculation with A. fumigatus conidia. Mice were infected with A. fumigatus via o.p. aspiration (107 organisms). As a positive control for susceptibility, mice underwent neutrophil depletion by i.p. injection of antibody clone RB6-8C5 (0.6 mg per mouse) 1 day prior to infection. (A) Survival and (B) weight loss of the animals were monitored for 4 days. (C) Fungal burden measured by primers and probes specifically detecting 18s RNA on day 2 after infection and cytokine gene expression in the lungs were determined by real-time RT-PCR.
