The role of MAVS aggregation in its function in mediating mitophagy. (a) HeLa cells were transfected with the indicated plasmids. Thirty-six hours after transfection, the total protein was extracted and subjected to immunoblotting analyses with the indicated antibodies. (b) HeLa cells were co-transfected with GFP-LC3 (green) together with wild-type Flag-MAVS or its aggregation mutant forms (red). Twenty-four hours after transfection, the cells were fixed, stained by the anti-Flag antibody and imaged by confocal microscopy. (c) The numbers of GFP-LC3 dots were quantified in (b) (mean±s.d. of ⩾50 cells). (d) HEK293 cells were transfected with the indicated plasmids.
