Effect of photoactivatable allosteric compounds on stimulation of agonist-induced [35S]GTPγS binding to the CB1 receptor. Dose-response curves for CP55,940-induced [35S]GTPγS binding to HEK293 cell membranes expressing the CB1 receptor in the absence and presence of 0.32 µM and 10 µM 26, 27, 28, or 36b. Data are presented as specific binding of GTPγS binding. Nonspecific binding was determined in the presence of 10 µM unlabeled GTPγS. The level of GTPγS binding of mock-transfected samples (non-CBl mediated) was subtracted from each specific binding data point. Each data point represents the mean ± S.E. (error bars) of at least three independent experiments performed in duplicated.
