Confocal imaging of N87 Her2 expressing cells treated with anti-HER2 antibody fluorophore conjugate demonstrate that internalization is not affected by sortase mediated ligation. Her2 expressing N87 cells were treated either with the anti-HER2 HC-C antibody or with HC-C-Cy3 antibody conjugate generated using SrtA mediated ligation. The cells were treated for 45 minutes at 4 °C, washed, and incubated for 24 hours prior to fixation and imaging by confocal microscope.
