Effects of collagen α-5(IV) chain (Col4a5) knockdown on the acidification of intracellular vesicles in RAW264.7 macrophages. (A) Dextran-Oregon Green 488 was added to the culture of control (VC) cells and cells in which and Col4a5 had been knockdown (C51, C52 and C53). After 1 and 5 h, the fluorescence was measured at the excitation wavelength 490 and 440 nm and converted to pH values via standard curve fitting. (B) Mycobacterium bovis Calmette-Guérin (BCG)-FITC was added to the culture of VC, C51, C52 and C53 macrophage cell lines. After 2 and 5 h, the fluorescence was measured at the excitation wavelength of 490 and 440 nm and converted to pH values via standard curve fitting. *P<0.05 compared with VC.
