PI3K inhibition induces airway dilation of small airways despite desensitization of the β₂‐adrenoceptors. hPCLS from four separate donors 79–81 slices/condition, total of 160 slices, were incubated in the presence or absence of 50 nM salmeterol (SALM; 24 h) to induce desensitization of the β₂‐adrenoceptor. After washout and treatment with carbachol, isoprenaline (ISO)‐induced airway dilation was significantly attenuated by salmeterol pretreatment. CAL‐101‐induced airway dilation, however, was indistinguishable from control pretreatment and was enhanced compared with the previous treatment of β agonist‐desensitized airways.
