The activities of the purified shikimate kinase was determined by coupling the release of ADP from the shikimate kinase-catalyzed reaction to the oxidation of NADH using pyruvate kinase and lactate dehydrogenase as coupling enzymes. Shikimate-dependent oxidation of NADH was monitored at A₃₄₀ (ε = 6,200 M⁻¹cm⁻¹) at 25 °C. The data are from three experimental replicates.
