A. PC3, LNCaP, DU145, HeLa, A549, PANC-1 and MiaPaCa-2 cells were transfected with control siRNA, and siRNAs to IKKα and IKKβ as indicated. The cells were lysed 48 hrs after transfection and the levels of IKKα, IKKβ, and β-actin and endogenous phosphorylation of S6K and Akt were determined by immunoblotting with the indicated antibodies. The experiments were carried out on three separate occasions. B. PC3 cells were transfected with siRNA control, siRNA-IKKα-2, or siRNA-IKKα-3, and lysed 48 hrs after transfection. Levels of the indicated proteins or phospho-proteins were detected by antibodies used in Figure 1A.
